For the following syntax in a code :

    while file.readline().startswith("#"):
        continue

Do i need to use "continue"? Or is there any other syntax for the same?

while file.readline().startswith("#"):
    pass

The pass statement does nothing. It can be used when a statement is required syntactically but the program requires no action.
